This research aims to model the unknown link between Coenzyme A (CoA) biosynthesis deficiency and increased arrhythmia risk, which is observed in patients with mutations like PPCS that can lead to dilated cardiomyopathy. The objectives are to develop a detailed model of cardiomyocyte electrophysiology incorporating CoA synthesis and energetics, determine how CoA deficiency modifies the cardiac action potential, and use 3D patient-specific simulations to investigate the combined effects of subcellular changes and geometric abnormalities on arrhythmia generation.

Our goal is to develop, test, and quality-assure tools that can accurately predict disease progression in individual patients, offering insights into potential future treatments and allowing for optimal intervention to prevent further disease development.
